Myostatin Inhibitors Compared: Follistatin-344, ACE-031 and ACE-083
Myostatin is one of the clearest examples of a single protein controlling tissue size. That has made it a long-standing target in muscle biology, and it explains why several quite different inhibitors appear in research catalogues.
What myostatin does
Myostatin (GDF-8) is a TGF-β family protein made mainly by skeletal muscle. It acts as a brake on muscle growth. Animals with natural loss-of-function mutations — the "double-muscled" cattle breeds, for example — have markedly increased muscle mass.
Mature myostatin binds activin type II receptors (mainly ActRIIB) and signals through SMAD2/3 to restrain the growth and differentiation of muscle precursor cells.
Three ways to block it
| Compound | Type | How it blocks myostatin |
|---|---|---|
| Follistatin-344 | Natural binding protein | Wraps around myostatin and activins, preventing receptor binding |
| ACE-031 | Receptor decoy (ActRIIB-Fc) | A soluble copy of the receptor soaks up ligands before they reach cells |
| ACE-083 | Follistatin-Fc fusion | Follistatin-based, engineered to act locally where it is placed |
Why selectivity matters
None of these is perfectly specific. Follistatin and ActRIIB decoys also bind activins and GDF-11, which have roles well beyond muscle. Clinical development of ACE-031 stopped early after unexpected vascular-related findings, which are thought to come from these broader binding effects. ACE-083's local design was one attempt to limit systemic exposure.
That is the main lesson from this field: blocking one pathway node often affects several biological systems at once.

Comparing listings
- These are proteins, not short peptides. They are much harder to make, so identity and activity data matter. Ask for SDS-PAGE and mass data, not just HPLC.
- Check the expression system and form — for example, whether follistatin is the 344 or 315 isoform.
- Compare price per microgram or milligram on each compound's comparison page. Typical quantities are small.
Summary
Follistatin, ACE-031 and ACE-083 all block myostatin signalling, but by different means and with different selectivity. PeptideCompare provides no dosing or usage guidance, and nothing here is for human or veterinary use.
